Incorporation of [3H]thymidine in mouse spleen.
نویسنده
چکیده
Mice were given 4 injections of [H]thymidine at 2-hourly intervals and killed at times varying from 1 h to 12 days after the last injection. Autoradiographs of spleen, oesophagus and heart were prepared by the stripping-film technique and exposed for 21, 189 and 367 days. Groups of lymphoid cells surrounding trabeculae were heavily labelled, the decline in grain count suggesting division of these cells within 24 h. The appearance of large numbers of labelled nucleated cells in venules suggests that cells leave the spleen through the lining of the venules. The cells in germinal centres were preponderantly weakly labelled and spread into the white pulp after 2 days. It is suggested that the weak labelling indicates turnover of DNA rather than pre-mitotic synthesis. Megacaryocytes were unlabelled 1 h after a single injection of [H]thymidine, nearly all being labelled from 1 to 8 days after the last of 4 injections. The grain count declined steadily with increasing interval between injection and fixation.

[Methyl-14C]-choline incorporation into nude mice bearing tumor xenografts-correlation with [methyl-3H]-thymidine: A pilot study
Background: Many in vitro studies suggested that choline incorporation into many types of tumors is related to cell proliferation. Whether in vivo choline incorporation is related also to cell proliferation or not was my question. Therefore, the aim of this pilot study was to investigate the relationship of in vivo [methyl- 14C]-choline incorporation and in vivo [methyl-3H]- thymidine incorpora...
متن کاملMitogenic activity of staphylococcal peptidoglycan.
Staphylococcus aureus peptidoglycan displayed a marked dose-dependent mitogenic activity for mouse splenocytes and human peripheral blood lymphocytes in vitro, as measured by increased [3H]thymidine incorporation. Similarly it was mitogenic for athymic nude mouse spleen cells, whereas no blastogenic effect was observed in T cell-enriched and B cell-depleted mouse lymphocyte cultures. These data...
متن کاملStimulation of DNA synthesis in vitro in mouse thymic cells by bone marrow preparation.
Some characteristics of a bone marrow factor were ascertained, on the basis of its ability to increase 3H-thymidine incorporation into thymic cells in vitro. It was found that the factor was not detectable in kidney or spleen preparations derived from the same mice from which active bone marrow preparations were obtained. The activity of the factor was reduced shortly after total-body irradiati...
متن کاملEffect of platinum and palladium salts on thymidine incorporation into DNA of rat tissues.
The intraperitoneal administration of PtCl4 or Pd(NO3)2 at levels of 28 or 56 mumole/kg body weight decreased the thymidine incorporation into DNA of spleen, liver, kidney, and testis. Spleen was most sensitive to both the platinum and the palladium salt. In liver, DNA syntheses in parenchymal cells and stromal cells were about equally sensitive to PtCl4. In control rats, only 20-30% of the 3H ...
متن کاملEffect of C-reactive protein, PHA, PWM and choline phosphate in 3H-thymidine uptake of leukocytes of leprosy patients and normal individuals.
Autologous serum used in lymphocyte cultures in transformation studies for the examination of cell-mediated immunity in leprosy patients has been shown to contain substance(s) which depressed the 3H-thymidine incorporation into DNA in phytohemagglutinin (PHA) stimulation of leukocytes (4.27). Initial attempts at physical and chemical characterization of the serum factor have been made by Bulloc...
متن کاملTissue pharmacokinetics and inhibition of DNA synthesis in mice treated with sporamycin.
Examination of blood and organ concentrations of sporamycin in normal mice showed a rapid decrease of sporamycin from peripheral blood and a high level of sporamycin in urine 10 minutes after intravenous injection of the antibiotic. At the same time, the highest level was found in the kidneys and low levels were found in the lungs and spleen. When sporamycin was added to mouse organ homogenate ...
